Optimization of Ultrasonic-Enhanced Extraction of Tobacco Total Polysaccharides
Abstract
In this study, the condition of ultrasound enhanced extraction of tobacco polysaccharides from tobacco leaves was optimized using response surface methodology (RSM). The relationship between the rate of polysaccharides and main process conditions including ultrasonic temperature (X1), time (X2) and solid-liquid ratio (X3) was modeled using a three-level three-factor BBD design based on various factors. Moreover the established model was analyzed by RSM to obtain the optimum extraction conditions. The result of analysis showed that X2, X3, X22, X32, X1X3 possess significant impact on the ultrasound enhanced extraction rate results. The optimal process conditions were X1 of 55℃, X2 of 45min, and X3 of 35:1, and the extraction yield reached 14.68% (n=3) under these conditions. The results provide a reference for extraction of tobacco polysaccharides, and lay a foundation for future research on tobacco
